  • 1.  High fat diet in vitro??

    Hello fellow Lipidologists,

    Has anyone simulated a high fat diet in vitro? I'd like to "fatten up" some immune cells and test the impact of "high fat" on immune function. 

    My plan is to titrate triolein and stain for lipid droplets as a quick and dirty way to see if the treatment is working. Is there is a better way?

    Typically we use a mix of fatty acids (palmitate and oleate) conjugated to albumin. We then use bodipy or oil red o to ensure lipid droplet formation. 

    Another option that we've used is media transfer from adipocytes after inducing lipolysis (CL-316,243). This is slightly more efficient. 

    Both methods lead to lipid droplet formation in hepatocyte cell lines in 24-48hrs.
